Combination of 5-fluorouracil and Lactic Acid Bacteria Induces Apoptosis Synergistically in HT-29 Colon Cancer Cells

Abstract
5-fluorouracil (5-FU) is one of the widely used chemo-therapeutic drugs targeting various cancer cells. A successful chemotreatment requires overcoming several obstacles including intrinsically drugchemoreisitant cancer malignancy. In the present study, HT-29 colon cancer cells were markedly sensitized to apoptosis by both 5-FU and lactic acid bacteria (LAB), compared to the LAB treatment alone. The precise mechanism by which LAB may inhibit the growth of colon cancer are presently unknown. It has been reported that treatment of HT-29 colon cancer cells with combined 5-FU and LAB markedly reduced tumor cell viability, in contrast to the results for LAB alone. Consequently, combination of 5-FU and LAB increased significantly activation of AMP-activated protein kinase (AMPK) that has been implicated in cancer development and control. These results suggest that the combination of 5-FU and LAB exert a novel chemotherapeutic effect in colon cancer cells.
